Title: Imaging and molecular radiotherapy of hypoxic tumors
Abstract: Hypoxia is a physiological state in which arises when the blood supply to a tissue diminishes. Hypoxic cells are alive, but their reduced cytoplasmic oxygen concentrations induce a range of changes in their biochemistry and molecular biology. Failure to reverse the ischaemic condition results in cellular anoxia and necrosis. Hypoxia increases resistance to radiotherapy and chemotherapy, up-regulates glycolysis and protein expression, and promotes angiogenesiscan. It can occur in most tissues, and is at play in many pathologies, including solid tumors, infections, inflammations, stroke, myocardial ischaemia, diabetes and arthritis.
Aspects of hypoxia-related chemistry, biochemistry and molecular biology will be presented as a background to a look at the development of agents for imaging of hypoxia in cancer. Recent approaches to improve the contrast of 2-nitroimidazoles, and their potential for use as molecular (radio)therapeutics, will be discussed.
