Title:     Design of the radionuclidic "toolbox" for immuno-PET imaging and novel drugs development.

Abstract: Immuno-PET imaging became effective tool for cancer diagnostic and new drugs screening [1, 2].

The main purpose of the project is to produce four PET radionuclides with different half-lives, which  match the wide range of biological half-lives of biomolecules. These radionuclides are  68Ga (t1/2 1.13 h), 44Sc(t1/2 3.97 h), 90Nb (t1/2 14.6 h) and 89Zr(t1/2 78.41 h). 68Ga and 44Sc would be ideally suitable for imaging with peptides and nanobodies with a molecular weight of less than 10 KDa and biological half-lives of hours. 90Nb and 89Zr will be suitable candidates for imaging of intact antibodies and antibody fragments with biological half-lives of days. Pairs of radionuclides can be also applied for a cross-reference study. Biomolecules will be labeled with 68Ga and 44Sc via DOTA-based chelators, while desferal (Df) will be used for labeling with 90Nb and 89Zr. Synthesis of universal chelators for all proposed radionuclides will be discussed.

Production strategies of all four radionuclides via direct (p,n) reactions , indirect (generators) and multinuclide production via high energy proton irradiations as well radiochemical separation strategies and generator design will be discussed.

Additionally, I would also like to initiate a discussion on other projects. In particular, irradiation of thorium targets for production of alpha-emitters for therapy (225Ac, 230U and 223Ra) and other medically- related fission products (103Ru, 111Ag, 117mSn etc.). Other projects will be focused on systematic evaluation of chemistry for medically-related radionuclides from the 5th and 15th groups of the periodic table (90Nb, 72,74As, 213Bi, 119Sb etc.).
