Title:     IsoSiM Student Seminars: "89Zr/177Lu-chelation with H4pypa and H4pypa-(linker)-Panitumumab"

Abstract: Both 177Lu and 89Zr have favorably long half-lives for labeling immunoconjugate (~6.6 days and ~3.3 days, respectively). However, due to the temperature- and pH-sensitive antibody, the radiolabeling is limited to <40 degC, pH = 6 – 7. Herein, H4pypa, a nonadentate non-macrocyclic chelator, is presented as a promising chelating agent for both radionuclides for antibody labeling.
The first part of the presentation involved the investigation of the chelation properties of H4pypa with 89Zr. Radiolabeling and different challenge studies would be discussed. 
The second part would focus on the radiochemical studies of two H4pypa-Panitumumab conjugates with 177Lu. H4pypa has previously demonstrated high affinity for 177Lu, however, there has not been any antibody-conjugate evaluated. Therefore, two N-isothiocyanate functionalized H4pypa with different linkers were developed, conjugated to the monoclonal antibody Panitumumab and evaluated for their chelation with 177Lu.
